The Science:

Why Oncolytic Viruses

Oncolytic viruses are naturally occurring or engineered viruses that preferentially infect and replicate inside cancer cells while largely sparing healthy tissue. Cancer cells are unusually vulnerable to these viruses: the same genetic changes that let a tumor grow uncontrollably —  such as impaired antiviral (interferon) defenses and hyperactive growth-signaling pathways — also make the tumor an ideal host for viral replication. 

The therapy works through two complementary mechanisms:

Direct tumor destruction (oncolysis) — where the virus multiplies inside cancer cells until they rupture, killing the tumor cell and releasing new viral particles that go on to infect neighboring cancer cells; and Immune activation — as tumor cells break apart, they release tumor antigens and danger signals into the surrounding tissue. This converts an immunologically "cold," hidden tumor into a "hot," visible one — effectively vaccinating the patient against their own cancer and recruiting the immune system to attack tumor cells throughout the body, including sites the virus never reached.

 

This dual action is what makes oncolytic viruses so compelling. They are not only a direct cytotoxic agent but also an in situ immunotherapy, and they can be engineered to carry therapeutic payloads that further amplify the anti-tumor response. The class is clinically validated — an oncolytic virus therapy has already earned regulatory approval in major markets — and the field is now advancing rapidly across many tumor types.

At Barkan Health, we select our specific viral strain based on our own preclinical models, matching the right platform and engineering strategy to the biology of our target indication.
